- 质粒简介: pCold ProS2 DNA 是一种融合的冷休克表达载体,使用来源于革兰氏阴性菌Myxococcus xanthus中的Protein S作为可溶性标签。这种Protein S存在于粘细菌的孢子衣中,由173个氨基酸残基组成,是一种非常稳定的可溶性蛋白质。将两个Protein S的N-末端结构域串联,再结合ProS2 Tag的作用,就能使融合目的蛋白的稳定性和可溶性得到提高。此外,由于目的蛋白和ProS2-Tag的相互作用性很低,因此可以将二者有效分离。pCold ProS2 DNA Vector在cspA启动子的下游插入了5’非编码区(5’-UTR)、翻译增强元件(TEE)、His-Tag序列、ProS2-Tag和多克隆位点(MCS)。在ProS2-Tag和多克隆位点(MCS)之间主要是HRV3C Protease、Thrombin和Factor Xa的切断位点,用来消除融合蛋白质的标签。此外,在cspA启动子下游还插入了lac operator,可以严格调控目的基因的表达。由于本制品的启动子是大肠杆菌来源的,所以大部分大肠杆菌都可以作为表达宿主使用。总之,pCold ProS2 DNAVector提供的冷休克技术,能够促进蛋白质的正确折叠,增加目的蛋白质的可溶性,对于一些棘手的目的蛋白质的表达是一个很好的选择。
